Chromosome breakage by 1-methyl-2-benzylhydrazine in mouse cancer cells.

چکیده

The induction of chromatid translocations by l-methyl-2-benzylhydrazine was studied in a number of malignant and normal mammalian cell types and in narcissus root cells. In three different mouse ascites tumors (Ehrlich, 2BF, and LI210) treated in vivo, translocations were induced. In the Ehrlich tumor, they were induced with unusually high frequency, involving, in some cells, a majority of the chromosomes. None were found in the Ehrlich tumor or HeLa cells in vitro, nor were any found in untreated controls. No translocations were observed in treated or untreated normal tissues in vivo (mouse bone marrow and spleen, narcissus roots) or in vitro (human lymphocytes, mouse spleen).

Metabolism of an antineoplastic methylhydrazine derivative in a P815 mouse neoplasm.

Ten normally occurring methylated trace bases of cytoplasmic RNA extracted from P815 ascites cells were identified as thymine, 3-methylcytosine, 5-methylcytosine, jV2-methylguanine, JV2, A^-dimethylguanine, 1-methylguanine, 7-methylguanine, 1-methyladenine, 6-methyladenine, and jV*,yV6-dimethyladenine. When the carcinostatic drug, l-methyl-2-(p-isopropylcarbamoyl)benzylhydrazine hydrochloride (...

Cells deficient in DNA polymerase beta are hypersensitive to alkylating agent-induced apoptosis and chromosomal breakage.

DNA polymerase beta (beta-pol), which is involved in base excision repair, was investigated for its role in protection of cells against various genotoxic agents and cytostatic drugs using beta-pol knockout mouse fibroblasts. We show that cells lacking beta-pol are highly sensitive to induction of apoptosis and chromosomal breakage by methylating agents, such as N-methyl-N'-nitro-N-nitrosoguanid...

I-38: Chromosome Instability in The Cleavage Stage Embryo

Recently, we demonstrated chromosome instability (CIN) in human cleavage stage embryogenesis following in vitro fertilization (IVF). CIN not necessarily undermines normal human development (i.e. when remaining normal diploid blastomeres develop the embryo proper), however it can spark a spectrum of conditions, including loss of conception, genetic disease and genetic variation development. To s...

Chromosome breakage induced by thymidylate stress in thymidylate synthase-negative mutants of mouse FM3A cells.

In thymidylate synthase-negative mutants of mouse FM3A cells, thymidine starvation rapidly decreased mitotic activity and resulted in cell death (thymineless death). When the thymidine starvation was reversed by an addition of thymidine, mitotic activity was recovered, but the majority of mitotic cells exhibited extensive chromosome aberrations, including chromatid breaks, chromatid exchanges, ...

Loss of DNA Polymerase Z Causes Chromosomal Instability in Mammalian Cells

Rev3L encodes the catalytic subunit of DNA polymerase Z (pol Z) in mammalian cells. In yeast, pol Z helps cells bypass sites of DNA damage that can block replication enzymes. Targeted disruption of the mouse Rev3L gene causes lethality midway through embryonic gestation, and Rev3L / mouse embryonic fibroblasts (MEFs) remain in a quiescent state in culture. This suggests that pol Z may be necess...
